1. Basic principles

However, the animal's inquiry characteristics in turn cause it to generate motives for activities in the central region (including exercise, drinking or eating), thus creating a state of conflict. Anxiolytics (such as diazepam) can relieve this state of conflict by showing that the dose that does not change the activity count significantly increases the number of times the rat enters the central region of the open box and the time spent in the central region (Stefanski method) or increases The drinking time of the rats in the central area (Stout-weiss method), the anxiety agent (such as FG7142) significantly reduced the drinking time.
Second, the operation steps
Stefanski method: the experimental device is a circular open box with a diameter of 80cm and a height of 30cm; the center of the bottom of the open box is divided into two inner and outer circles with diameters of 35cm and 50cm respectively, and the inner ring contains the central area; the side near the bottom Three photocells are symmetrically mounted on the wall, and the activity of the mouse can be observed through the television. The experiment was performed under low light illumination, 65 dB of white noise (background sound). Male Wistar rats were used for experimentation and weighed 200-240 g. The rats were placed in the center of the open box, and the total activity count of the rats within 10 minutes, the number of times of entering the outer ring of the open box (about 15 cm from the periphery of the open box to the central area) and the time spent in the central area of ​​the open box were observed. .
Stout-weiss method: (the open field drink test, OFDT), the experiment uses a 36cm*36cm*36cm transparent plexiglass box, the bottom of which is painted black, the top is open (this experimental instrument can be soft by Shanghai Xinxin Information Technology Co., Ltd. provides). The middle of the box hangs a water bottle upside down, the mouth of the bottle is 10cm from the ground; there is a 25w incandescent lamp 60cm above the ground for illumination. The rats were stroked for 1 min every day at least three days before the experiment to reduce the effect of non-specific stress on the experiment; only 1 hour (3:00-4:00 pm) of drinking water per day for the first 3 days of the experiment. During the experiment, the rats were placed in the center of the open box, and the incubation period, drinking time (main index), the number of water bottles and the number of standings of the rats were observed within 10 minutes.
III. Notes and evaluation
(1) After each rat experiment, the test chamber was thoroughly cleaned with a 5% aqueous solution of acetic acid to prevent olfactory suggestion from being transmitted to the next rat.
(2) Because of its simple equipment and simple operation, the open box experiment is very common in behavioral research and is used to evaluate the comprehensive behavior of animal activity. As an anxiety model, it has only been developed in recent years, and the experimental data obtained is limited. Therefore, its stability and reliability have yet to be further verified, and it is best to evaluate the effects of drugs in combination with other anxiety models. It is worth noting that although buspirone is not sensitive on many other models, 0.3 mg/kg has an anxiolytic effect in this experiment.
