There are many types of microtome, including medicinal slicer, equipment slicer, food slicer, tissue slicer and so on. Tissue microtome is mainly a device for pathological section analysis of human tissues and animal and plant tissues. Tissue microtome is generally divided into: rotary microtome and cryostat. Rotary slicer: It is a slicing action by turning the hand wheel. The wax block is mounted in a metal holder that can move up and down in the groove, and the micro-motion spiral advances to cut the flat slice. Some rotary microtome machines have three knobs and a fastening knob on the nose to deflect and tighten in all directions, which is convenient for adjusting the cut surface of the wax block. The cutting angle of the slicing knife can be adjusted (slicing knife tilt). Since a large and large slicing knife is used on the microtome, generally no vibration occurs in the cutting of the hard tissue. The slice thickness can be adjusted to any thickness between 1-30 microns using a knob, each gradient being 1 or 2 microns. The advantage of the rotary slicer is that the body is heavier, so the rocking slicer is stable, which is very suitable for cutting paraffin sections. It can ideally cut continuous slices and cut large tissue blocks. Cryosection is a technical method of freezing fresh tissue specimens to produce a certain hardness by physical cooling. Compared with paraffin sections, cryopreservation does not require dehydration and entrapment, so the speed of tableting is a good method for clinicians to provide pathological diagnosis. In addition, since frozen sections are unfixed fresh tissue, cryosections are also an ideal method for fat staining, enzymatic histochemical staining, and some immunohistochemical staining and in situ molecular hybridization. The deficiency of frozen sections is that the morphology of the tissue cells is slightly inferior to that of the paraffin sections. It is widely used in hospitals, medical schools, forensic science, life sciences, health and epidemic prevention, public security forensics, agricultural colleges, scientific research institutions and other units to produce pathological and biological tissue sections, animal and plant research units for pathological diagnosis, analysis and research. The microtome has a rotary and slide-away microtome for optical microscopy. For ultra-thin microtome for electron microscopy, use ultra-thin sections with glass or diamond knives. The material of the device is mechanically propelled and thermally expanded. For the cryosection method, there are a cryostat, a cryostat, and a frozen ultramicrotome.
