Imatinib is a Kinase Inhibitor. The mechanism of action of imatinib is as a Protein Kinase Inhibitor.
Imatinib, sold under the brand names Gleevec among others, is a chemotherapy medication used to treat cancer. Specifically, it is used for chronic myelogenous leukemia (CML) and acute lymphocytic leukemia (ALL) that is Philadelphia chromosome-positive (Ph+) and certain types of gastrointestinal stromal tumors (GIST), systemic mastocytosis, and myelodysplastic syndrome. It is taken by mouth.
Common side effects include vomiting, diarrhea, muscle pain, headache, and rash. Severe side effects may include fluid retention, gastrointestinal bleeding, bone marrow suppression, liver problems, and heart failure. Use during pregnancy may result in harm to the baby. Imatinib works by stopping the Bcr-Abl tyrosine-kinase. This either slows growth or results in programmed cell death of certain type of cancer cells.
Medical uses
Imatinib is used to treat chronic myelogenous leukemia (CML), gastrointestinal stromal tumors (GISTs) and a number of other malignancies.
Imatinib Base is an antineoplastic agent that inhibits the Bcr-Abl fusion protein tyrosine kinase, an abnormal enzyme produced by chronic myeloid leukemia cells that contain the Philadelphia chromosome. Imatinib also inhibits the receptor tyrosine kinases for platelet-derived growth factor (PDGF) and stem cell factor (SCF)/c-kit; the SCF/c-kit receptor tyrosine kinase is activated in gastrointestinal stromal tumor (GIST). This agent inhibits proliferation and induces apoptosis in cells that overexpress these oncoproteins.
Imatinib is used for:
Treating certain types of leukemia, certain bone marrow diseases, a certain type of skin cancer, and certain intestinal tumors (eg, gastrointestinal stromal tumors ). It may also be used to prevent cancer from growing in patients after surgical removal of GIST. It is also used to treat mastocytosis (a buildup of large amounts of mast cells in certain parts of the body) or hypereosinophilic syndrome (a buildup of large amounts of eosinophils in the body).Â
Imatinib is a protein-tyrosine kinase inhibitor. It works by preventing the growth of cancer cells.
Side effects
The most common side effects include feeling sick (nausea), diarrhea, headaches, leg aches/cramps, fluid retention, visual disturbances, itchy rash, lowered resistance to infection, bruising or bleeding, loss of appetite;weight gain, reduced number of blood cells (neutropenia, thrombocytopenia, anemia), and edema. Although rare, restoration of hair color has been reported as well.Â
Dosage
The prescribed dose should be administered orally, with a meal and a large glass of water. Doses of 400 mg or 600 mg should be administered once daily, whereas a dose of 800 mg should be administered as 400 mg twice a day.
Treatment may be continued as long as there is no evidence of progressive disease or unacceptable toxicity.
